Abstract: Multiple sclerosis (MS) is an autoimmune disease that causes demyelination and neurodegeneration. A major unmet need in MS treatment is the development of therapies that restore immune tolerance to self-antigens. Our group have explored an innovative approach using phosphatidylserine liposomes that mimic apoptotic bodies and carry a myelin peptide. In an experimental autoimmune encephalomyelitis (EAE) model, these liposomes demonstrated therapeutic efficacy both before and after symptom onset, through clinically relevant administration routes such as intravenous injection. The mechanism involves expansion of regulatory T and B cells and suppression of MOG-specific inflammatory responses. This strategy offers a safe, versatile, and promising path toward clinical translation in MS.
